I try and keep them like this in between rides, just sitting on my stand. It makes a noticeable difference. Added benefit, if there's little fingers running around it'll keep them from sticking their fingers in spinning brake rotors.

Most systems are designed to encourage air to collect vertically, so if you're going to do this the brake levers should be in a fairly normal position, somewhere between 45 degrees and parallel to the ground.

Do it overnight yes. It allows for larger volume behind the pistons and in theory the bubbles will float up. Pro trick, take your wheel out and clamp down on a dummy rotor, rubber band and leave over night. In the morning, while holding the lever closed and removing the rubber band, use the rotor to push the pistons back into place and allow that pressure to extend the brake lever against your finger. Helps push any bubbles up. Shimanos are usually done with the bleed cup on over night.
